Specificity
Clone 1F18 is a ZooMAb® rabbit recombinant monoclonal antibody that detects β-NGF. It targets an epitope within 15 amino acids from the N-terminal region of mature β-NGF.
Immunogen
KLH-conjugated linear peptide corresponding to 15 amino acids from the N-terminal region of human β-NGF.
Application
Quality Control Testing
Evaluated by Western Blotting in Mouse heart tissue lysate.
Western Blotting Analysis: A 1:1,000 dilution of this antibody detected β-NGF in Mouse heart tissue lysate.
Tested applications
Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected β-NGF in lysates from Human brain tissue, HeLa cells, NCI-H460 cells, and Rat thyroid tissue.
Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected β-NGF in human brain and rat brain tissue sections.
Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected β-NGF in A431, HepG2 and NIH 3T3 cells..

Target description
Beta-nerve growth factor (UniProt: P01138; also known as β-NGF) is encoded by the NGF (also known as NGFB) gene (Gene ID: 4803) in human. β-NGF, an endogenous neurotrophic factor, is a homodimeric protein that is synthesized with a signal peptide (aa 1-18) and a propeptide (aa 19-121) that are subsequently cleaved off to generate the mature form (aa 122-241). Nerve growth factor is important for the development and maintenance of the sympathetic and sensory nervous systems. It serves as an extracellular ligand for the NTRK1 and NGF receptors to activate cellular signaling cascades to regulate neuronal proliferation, differentiation, and survival. However, it does not permeate the blood-brain barrier. Its precursor form (ProNGF) is reported to function as a ligand for the heterodimeric receptor formed by Sortilin Related VPS10 Domain Containing Receptor 2 (SORCS2) and NGF receptor that activates signaling cascade leading to inactivation of RAC1 and/or RAC2. In contrast to the mature form, ProNGF is known to promote neuronal apoptosis. N-glycosylation of ProNGF is reported to be essential for its processing, secretion, and construction of the tertiary fold of the β-NGF homodimer. Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.
Reconstitution
300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Storage and Stability
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
