General description
Glutamate receptor ionotropic, NMDA 2B (UniProt: Q00960; also known as GluN2B, Glutamate [NMDA] receptor subunit epsilon-2, N-methyl D-aspartate receptor subtype 2B, NMDAR2B, NR2B) is encoded by the Grin2b gene (Gene ID: 24410) in rat. NMDA receptor complexes are ligand-gated ion channels with high calcium permeability and voltage-dependent sensitivity to magnesium. They are involved in long-term potentiation and neuronal plasticity and regulate the number of nerve cells during development. However, over-stimulation NMDA receptors can lead to neuronal degeneration. NMDAR2B is a multi-pass membrane protein, which is synthesized with a signal peptide (aa 1-26), that is subsequently cleaved off to generate the mature form that contains two extracellular domains, three cytoplasmic domains, and four transmembrane domains. It is strongly expressed at birth and its levels increase until P6 to P20 and then begins to decline to the adult level. NMDAR2B is a component of the NMDA receptor complex that functions as heterotetrameric, ligand-gated ion channel with high calcium permeability and voltage-dependent sensitivity to magnesium. The channel activation requires binding of glutamate to the epsilon subunit, glycine binding to the zeta subunit, plus membrane depolarization to eliminate channel inhibition by Mg2+. Specificity
Clone 1C6.5C4 is a mouse monoclonal antibody that detects NMDAR2B. It targets an epitope within 136 amino acids from the C-terminal half.
Immunogen
His-tagged recombinant fragment corresponding to 136 amino acids from the C-terminal half of Rat NMDAR2B.
Application
Quality Control Testing
Evaluated by Western Blotting in Rat brain tissue lysate.
Western Blotting Analysis: A 1:500 dilution of this antibody detected NMDAR2B in Rat brain tissue lysate.
