Specificity
Clone 1E8 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects Translocator protein (PBR/TSPO). It targets an epitope within 14 amino acids from the C-terminal, third cytoplasmic domain.
Immunogen
KLH-conjugated linear peptide corresponding to 14 amino acids from the third cytoplasmic domain within the C-terminal region of human Translocator protein (PBR/TSPO).
Application
Quality Control Testing
Evaluated by Western Blotting in HeLa cell lysate.
Western Blotting Analysis: A 1:1,000 dilution of this antibody detected PBR/TSPO in HeLa cell lysate.
Tested Applications
Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected PBR/TSPO in U87-MG, and Human kidney tissue lysates.
Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected PBR/TSPO in Human colon tissue sections.
Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected PBR/TSPO in A431 cells.

Target description
Translocator protein (UniProt: P30536; also known as Mitochondrial benzodiazepine receptor, PKBS, Peripheral-type benzodiazepine receptor, PBR) is encoded by the TSPO (also known as BZRP, MBR) gene (Gene ID: 706) in human. TSPO is a multi-pass outer mitochondrial membrane protein with three mitochondrial intermembrane domain, five transmembrane domains, and three cytoplasmic domains. It is expressed in many tissues with highest expression observed in tissues that synthesize steroids. It plays an important role in translocating cholesterol from the cytoplasm into mitochondria, which is a rate limiting step in steroid synthesis. TSPO expression is also observed in the microglia in the central nervous system, and its expression is greatly increased upon microglial activation. It is considered as a sensitive biomarker of brain damage and neurodegeneration, particularly of inflammation and reactive gliosis. TSPO is shown to bind with voltage-dependent anion channel (VDAC) and the adenine nucleotide transporter (ANT). It can bind protoporphyrin IX and may play a role in the transport of porphyrins and heme. It promotes the transport of cholesterol across mitochondrial membranes and may play a role in lipid metabolism. However, it is apparently not required for steroid hormone biosynthesis. Two isoforms of TSPO are descried that are produced by alternative splicing. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
Reconstitution
0.3 mg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Storage and Stability
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
