Specificity
Clone 1E20 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects Programmed cell death protein 4 (PDCD4). It targets an epitope within 19 amino acids from the N-terminal half.
Immunogen
KLH-conjugated linear peptide corresponding to 19 amino acids from the N-terminal half of human Programmed cell death protein 4 (PDCD4).
Application
Quality Control Testing
Evaluated by Western Blotting in RAW264.7 cell lysate.
Western Blotting Analysis: A 1:3,000 dilution of this antibody detected PDCD4 in RAW264.7 cell lysate.
Tested applications
Western Blotting Analysis: A 1:3,000 dilution from a representative lot detected PDCD4 in human colon tissue lysate.
Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected PDCD4 in MCF-7 cells.
Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected PDCD4 in human prostate tissue sections.
Affinity Binding Assay: A representative lot of this antibody bound PDCD4 with a KD of 2.5 x 10-8 in an affinity binding assay.

Target description
Programmed cell death protein 4 (UniProt: Q53EL6; also known as Neoplastic transformation inhibitor protein, Nuclear antigen H731-like, Protein 197/15a, PDCD4) is encoded by the PDCD4 (also known as H731) gene (Gene ID: 27250) in human. PDCD4 is a tumor-suppressor protein that is shown to be upregulated during apoptosis. It affects transcription, translation, and signal transduction to suppress tumor growth. Its levels are upregulated in proliferative cells and lower levels are reported in lung cancer and colon carcinoma. Its levels are also down regulated in transitional cell carcinoma. PDCD4 is reported to shuttle between the nucleus and cytoplasm but is predominantly nuclear under normal growth conditions and when phosphorylated at serine 457. It contains two nuclear localization signal sequences (aa 58-64 and 241-250). PDCD4 can undergo phosphorylation at serine 67 by Ribosomal protein S6 kinase beta-1 (RPS6KB1) in response to mitogens, which promotes its proteasomal degradation. PDCD4 is known to undergo methylation at arginine 110 that enhances its interaction with elF4A. The methylated form of PDCD4 promotes tumor viability during nutrient deprivation, which allows tumors to grow more aggressively. This methylation can reinforce the interaction between the MA3 domains and eIF4A thereby forming a stronger interaction and may also make PDCD4 MA3 domains more accessible for interaction with eIF4A. Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.
Reconstitution
300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Storage and Stability
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
