Specificity
Clone 1E9 is a ZooMAb rabbit recombinant monoclonal antibody that specifically detects cAMP-specific 3′,5′-cyclic phosphodiesterase 4B (PDE4B). It targets an epitope within 22 amino acids from the C-terminal region.
Immunogen
KLH-conjugated linear peptide corresponding to 22 amino acids from the C-terminal region of human cAMP-specific 3′,5′-cyclic phosphodiesterase 4B (PDE4B).
Application
Anti-PDE4B, clone 1E9 ZooMAb, Cat. No. ZRB1583, is a recombinant Rabbit monoclonal antibody that specifically targets PDE4B and is tested for use in Affinity Binding Assay, Immunocytochemistry, Immunohistochemistry, and Western Blotting.
Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected PDE4B in lysates from HL-60, HT-29, and HepG2 cells and mouse brain tissue lysate.
Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected PDE4B in HepG2 cells.
Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected PDE4B in human placenta tissue sections.
Affinity Binding Assay: A representative lot of this antibody bound PDE4B peptide with a KD of 1.2 x 10-6 in an affinity binding assay.

Target description
cAMP-specific 3′,5′-cyclic phosphodiesterase 4B (UniProt: Q07343; also known as EC:3.1.4.53, DPDE4, PDE32) is encoded by the PDE4B (also known as DPDE4) gene (Gene ID: 5142) in human. Cyclic nucleotide phosphodiesterases (PDEs) play a critical role in the regulation of cAMP signaling. They degrade and inactivate cAMP and terminate its action. PDE4B is expressed in brain, heart, lung and skeletal muscle. It is also the predominant PDE subtype in monocytes and neutrophils. Two variants of PDE4B (PDE4B1 and PDE4B2) have been described in human and rat. Its cAMP binding site of PDE4B is localized in amino acids 406-410. PDE4B is shown to be crucial in the regulation of cardiac excitation-contraction coupling (ECC) and its inhibition increases the tendency of cardiac arrhythmias. It is reported to function in a confined compartment near the sarcolemma of cardiac myocytes and it controls subsarcolemmal cAMP but not the cytosolic cAMP. A reduction in PDE4B expression and activity has been observed during cardiac hypertrophy. Four isoforms of PDE4B have been described that are produced by alternative splicing. This ZooMAb recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.
Reconstitution
300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Storage and Stability
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
