Specificity
Clone 1E13 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects PDK1. It targets an epitope within 19 amino acids from the C-terminal half.
Immunogen
KLH-conjugated linear peptide corresponding o 19 amino acids from the C-terminal half of human PDK1.

Quality Control Testing
Evaluated by Western Blotting in NIH3T3 cell lysate.
Western Blotting Analysis: A 1:1,000 dilution of this antibody detected PDK1 in NIH3T3 cell lysate.
Tested applications
Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected PDK1 in Jurkat and PC12 cell lysates.
Affinity Binding Assay: A representative lot of this antibody bound recombinant Human PDK1 protein with a KD of 1.4 x 10-6 in an affinity binding assay.
Enzyme Immunoassay Analysis (ELISA): Serial dilutions from a representative lot detected PDK1 in ELISA application.

Target description
3-phosphoinositide-dependent protein kinase 1 (UniProt: O15530; also known as EC:2.7.11.1, hPDK1) is encoded by the PDPK1 (also known as PDK1) gene (Gene ID: 5170) in human. PDK-1 is a serine/threonine protein kinase that serves as a master kinase, phosphorylating and activating a subgroup of the AGC family of protein kinases. Its protein kinase domain is localized near the N-terminal (aa 82-342) and the pleckstrin homology domain (aa 459-550) resides near the C-terminal region. Its targets include Akt, p70 and p90 ribosomal protein S6 kinases, PKA, PKC, PAK-1, and serum and glucocorticoid-inducible kinase. It plays a key role in the transduction of signals from insulin by providing the activating phosphorylation to Akt 1, thus propagating the signal to downstream targets controlling cell proliferation and survival, as well as glucose and amino acid uptake and storage. It is reported to negatively regulate the TGF-β- induced signaling by modulating the association of SMAD3 and SMAD7 with TGF-β receptor and preventing the nuclear translocation of SMAD3 and SMAD4 as well as the translocation of SMAD7 from the nucleus to the cytoplasm in response to TGF-β. Its homodimerization is known to regulate its activity by maintaining the kinase in an autoinhibitory conformation. PDK-1 activity can be downregulated by NPRL2 that interferes with its phosphorylation at tyrosine 9, 373, and 376. NPRL2 also down-regulates its activity by interfering with tyrosine phosphorylation at the tyrosine 9, 373, and 376. The 14-3-3 protein theta (YWHAQ) also acts as its negative regulator by association with the residues surrounding the serine 241. Phosphorylation on serine 241 in the activation loop is required for its full activity. PDK-1 itself can autophosphorylate serine 241, leading to its own activation. Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.
Reconstitution
300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Storage and Stability
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
