Specificity
Clone 2H11 is a ZooMAb rabbit recombinant monoclonal antibody that specifically detects human Pericentrin. It targets an epitope within 132 amino acids from the N-terminal region.
Immunogen
GST/His-tagged recombinant fragment corresponding to 132 amino acids from the N-terminal region of human Pericentrin.
Application
Anti-Pericentrin, clone 2H11 ZooMAb, Cat. No. ZRB1318, a Rabbit monoclonal antibody that targets human Pericentrin and is tested in Affinity Binding Assay, Immunocytochemistry, Immunohistochemistry, and Western Blotting.
Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected Pericentrin in HeLa, A431, HUVEC and HepG2 cell lines.
Affinity Binding Assay: A representative lot of this antibody bound recombinant Pericentrin with a KD of 2.7 x 10-9 in an affinity binding assay.
Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected Pericentrin in HeLa and HEK293 cell lysates.

Target description
Pericentrin (UniProt: O95613; also known as Kendrin, Pericentrin-B) is encoded by the PCNT (also known as KIAA0402, PCNT2) gene (Gene ID: 5116) in human. Pericentrin is a highly conserved protein of the pericentriolar material and is an integral component of the filamentous matrix of the centrosome involved in the initial establishment of organized microtubule arrays in both mitosis and meiosis. The centrosomal localization of pericentrin is shown to be mediated by a pericentrosomal matrix targeting motif, known as PACT domain, in its C-terminal region. Pericentrin plays a role, together with DISC1, in the microtubule network formation. It is also reported to prevent premature centrosome splitting during interphase by inhibiting NEK2 kinase activity at the centrosome. It is also involved in retinal development by regulating photoreceptor ciliogenesis and photoreceptive outer segment formation and plays a role in ciliary development in olfactory receptor neurons. Pericentrin is shown to bind to calmodulin by a C-terminal binding site (aa 3195-3208) and is essential for its functions at centrosomes. During mitosis, pericentrin is cleaved to remove CDK5RAP2 that promotes centriole disengagement and subsequent centriole separation. Mutations in PCNT2 gene are known to cause microcephalic osteodysplastic primordial dwarfism that is characterized by microcephaly and short stature. Reconstitution
30 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Storage and Stability
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
