General description
Keratin, type I cytoskeletal 20 (UniProt P35900; also known as CK-20, Cytokeratin-20, K20, Keratin-20, Protein IT) is encoded by the KRT20 gene (Gene ID 54474) in human. Cytokeratin-20 (Keratin-20 or K20) belongs to a family of highly conserved epithelial-specific intermediate filament (IF) proteins that play an important role in the mechanical stability and integrity of epithelial cells and tissues. Each epithelium is characterized by its unique keratin expression pattern. All epithelial cells typically express at least one type-I and one type-II keratin, which form noncovalent heteropolymers. keratinocytes preferentially express K5/K14 or K1/K10 depending on their differentiation state in the epidermis, adult hepatocytes express K8/K18 exclusively, whereas intestinal epithelial cells express K8 as the major type II keratin with varying levels of the type I K18/K19/K20, depending on the differentiation state. Keratin 20 is a type I intermediate filament protein preferentially expressed in epithelia of the stomach, intestine, uterus, and bladder, as well as in skin Merkel cells. Because of its restricted tissue distribution, K20 is widely used as a marker to help assess the origin of metastatic tumors. K20 is phosphorylated on Ser13 during cellular apoptosis induction and tissue injury. PMA treatment is shown to induce cellular K20 Ser13 phosphorylation, implicating protein kinase C (PKC) as being one of the kinases that target the phosphorylation site.
Specificity
This rabbit polyclonal antibody detected upregulated cellular Keratin-20 Ser13 phosphorylation upon phosphatase inhibitors treatment. Target site sequence is highly homologous, but not 100% conserved, among human, murine and rat species.
