General description

Specificity

Clone 2B7 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects p38 MAP kinase phosphorylated on threonine 180 and tyrosine 182.

Immunogen

KLH-conjugated linear peptide corresponding to 14 amino acids surrounding phosphothreonine 180 and phosphotyrosine 182 of human p38 MAP kinase delta.

Application

Quality Control Testing

Evaluated by Western Blotting in lysate from HEK293 cells treated with Anisomycin.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected phospho-p38 MAPK (Thr180/Tyr182) in lysate from overnight serum starved HEK293 cells treated with Anisomycin (5 g/mL; 30 min.)

Tested Applications

Peptide Inhibition Assay:: Target band detection in lysate from HEK293 cells treated with Anisomycin was prevented by pre-blocking of a representative lot with the immunogen phosphopeptide, but not the corresponding non-phosphopeptide.

Flow Cytometry Analysis: 0.1 µg from a representative lot detected phospho-p38 MAPK (Thr180/Tyr182) in one million HEK293 cells treated with Anisomycin.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected phospho-p38 MAPK (Thr180/Tyr182) in Human colon cancer tissue sections.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Mitogen-activated protein kinase 13 (UniProt: O15264; also known as EC:2.7.11.24, MAP kinase 13, MAPK 13, Mitogen-activated protein kinase p38 Delta, MAP kinase p38 Delta, Stress-activated protein kinase 4) is encoded by the MAPK13 (also known as PRKM13, SAPK4) gene (Gene ID: 5603) in human. p38 is a serine/threonine kinase that is an essential component of the MAP kinase signal transduction pathway. It plays an important role in the cascades of cellular responses evoked by extracellular stimuli such as pro-inflammatory cytokines or physical stress leading to direct activation of transcription factors such as ELK1 and ATF2. Four splice variants of the p38 family have been identified and are known as p38α, p38β, p38γ, and p38δ. Of these, p38α and p38β are ubiquitously expressed while p38γ and p38δ are differentially expressed depending on tissue type. p38δ expression is observed in testes, pancreas, small intestine, lung, and kidney. It is also abundant in macrophages and is also detected in neutrophils, CD4+ T-cells, and endothelial cells. It phosphorylates a broad range of downstream proteins. Its protein kinase domain is localized in amino acids 25-308. It is fully activated following Dual® phosphorylation on threonine 180 and tyrosine 182 by MKK3, MKK4, MKK6, and MKK7. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, Affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
