Specificity
Clone 1H16 is a Rabbit recombinant monoclonal antibody that specifically detects Protein arginine N-methyltransferase 5 (PRMT5). It targets an epitope within 24 amino acids from the N-terminal half.
Immunogen
KLH-conjugate linear peptide corresponding to 24 amino acids from the N-terminal half of human Protein arginine N-methyltransferase 5 (PRMT5).
Application
Quality Control Testing
Evaluated by Western Blotting in Raji cell lysate.
Western Blotting Analysis: A 1:1,000 dilution of this antibody detected PRMT5 in Raji cell lysate.
Tested applications
Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected PRMT5 in lysates from Jurkat cells and Mouse brain and Rat brain tissue.
Affinity Binding Assay: A representative lot of this antibody bound PRMT5 with a KD of 5.3 x 10-7 in an affinity binding assay.
Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected PRMT5 in human prostate tissue sections.
Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected PRMT5 in RAW264.7 cells.

Target description
Protein arginine N-methyltransferase 5 (UniProt: O14744; also known as EC: 2.2.2.320, 72 kDa ICln-binding protein, Histone-arginine N-methyltransferase PRMT5, Jak-binding protein 1, Shk1 kinase-binding protein 1 homolog, SKB1 homolog, SKB1Hs) is encoded by the PRMTS (also known as HRMT1L5, IBP72, JBP1, SKB1) gene (Gene ID: 10419) in human. PRMT5 is a ubiquitously distributed protein that catalyzes the formation of omega-N monomethylarginine (MMA) and symmetrical dimethylarginine (sDMA), with a preference for the formation of MMA. It specifically mediates the symmetrical dimethylation of arginine residues in the small nuclear ribonucleoproteins Sm D1 (SNRPD1) and Sm D3 (SNRPD3); such methylation being required for the assembly and biogenesis of snRNP core particles. It is also reported to methylate arginine 8 on histone H3 that can repress transcription. It can attenuate EGF signaling through the MAPK1/MAPK3 pathway acting at 2 levels. First, monomethylates EGFR, which enhances tyrosine 1197 phosphorylation and PTPN6 recruitment, eventually leading to reduced SOS1 phosphorylation. Second, it methylates RAF1 and probably BRAF, hence destabilizing these 2 signaling proteins and reducing their catalytic activity. Five isoforms of PRMT5 have been reported that are produced by alternative splicing. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.
Reconstitution
300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Storage and Stability
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
