Specificity
Clone 1G23 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects Polypyrimidine tract-binding protein 1 (PTBP1). It targets an epitope within 18 amino acids from the N-terminal half.
Immunogen
KLH-conjugated linear peptide corresponding to 18 amino acids from the N-terminal half of human Polypyrimidine tract-binding protein 1 (PTBP1/PTB).
Application
Quality Control Testing
Evaluated by Western Blotting in NIH3T3 cell lysate.
Western Blotting Analysis: A 1:1,000 dilution of this antibody detected PTBP1/PTB in NIH3T3 cell lysate.
Tested Applications
Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected PTBP1/PTB in L6 cell lysate.
Affinity Binding Assay: A representative lot of this antibody bound PTBP1/PTB peptide with a KD of 6.7 x 10-6 in an affinity binding assay.
Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected PTBP1/PTB in human small intestine tissue sections.

Target description
Polypyrimidine tract-binding protein 1 (UniProt: P26599; also known as PTB, 57 kDa RNA-binding protein PPTB-1, Heterogeneous nuclear ribonucleoprotein I, hnRNP I) is encoded by the PTBP1 (also known as PTB) gene (Gene ID: 5725) in human. PTB is a monomeric, sequence-specific, RNA binding protein that is involved in multiple aspects of messenger RNA metabolism, including the repression of alternative exons. It contains four RNA binding domains (RBDs) with each bound to a CUCUCU oligonucleotide. Three linker amino acid sequences separate the RBDs along the polypeptide sequence. Each RBD binds RNA with a different binding specificity. In PTB RBD3 and RBD4 interact, resulting in an antiparallel orientation of their bound RNAs. Hence, PTB induces RNA looping when bound to two separated pyrimidine tracts within the same RNA. PTB plays a role in pre-mRNA splicing and in the regulation of alternative splicing events. It activates exon skipping of its own pre-mRNA during muscle cell differentiation. It is reported to bind to polypyrimidine-rich controlling element (PCE) of CFTR and promote exon skipping of CFTR exon 9, thereby antagonizing TIA1 and its role in exon inclusion of CFTR exon 9. It is reported to be involved in the splicing of pyruvate kinase PKM by binding repressively to a polypyrimidine tract flanking PKM exon 9, inhibiting exon 9 inclusion and resulting in exon 10 inclusion and production of the PKM M2 isoform. isoforms of PTB have been described that are produced by alternative splicing. Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
Reconstitution
0.3 mg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Storage and Stability
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
