Specificity

Clone 2K12 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects RET. It targets an epitope within 18 amino acids from the cytoplasmic domain in the C-terminal half. Reacts with both isoforms.

KLH-conjugated linear peptide corresponding to 18 amino acids from the cytoplasmic domain in the C-terminal half of human Proto-oncogene tyrosine-protein kinase receptor Ret (RET).

Application

Quality Control Testing

Evaluated by Western Blotting in Neuro2A cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected RET in Neuro2A cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected RET in SH-SY5Y and SK-N-SH cell lysates.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected RET in SH-SY5Y cells.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected RET in human colon tissue sections.

Target description

Proto-oncogene tyrosine-protein kinase receptor Ret (UniProt: P07949; also known as EC: 2.7.10.1, Cadherin family member 12, Proto-oncogene c-Ret) is encoded by the RET (also known as CDHF12, CDHR16, PTC, RET51) gene (Gene ID: 5979) in human. RET is a single-pass type I membrane protein that is synthesized with a signal peptide (aa 1-28), which is subsequently cleaved off to produce the mature protein that contains an extracellular domain (aa 29-635), a transmembrane domain (aa 636-657), and a cytoplasmic domain (aa 658-1114). RET is a receptor tyrosine-protein kinase that is involved in numerous cellular mechanisms including cell proliferation, neuronal navigation, cell migration, and cell differentiation. It is also involved in orchestration intestine organogenesis, development of enteric nervous system, and renal organogenesis during embryonic life. It also promotes the formation of Peyer′s patch-like structures, a major component of the gut-associated lymphoid tissue. RET is shown to be active in the absence of ligand, triggering apoptosis through a mechanism that requires receptor intracellular caspase cleavage. In the presence of the ligand GDNF in somatotrophs, it promotes survival and down regulates growth hormone production. However, in the absence of GDNF, it triggers apoptosis. Its protein kinase domain is localized in amino acids 724-1016. RET is auto-phosphorylated on C-terminal tyrosine residues upon ligand stimulation. The phosphorylated form is known to interact with phospholipase 1, and growth factor receptor bound protein 7 (Grb7). It undergoes dephosphorylation on tyrosine residues 905, 1015, and 1062 by protein tyrosine phosphatase receptor type J (PTPRJ). Mutations in RET gene have been linked to multiple types of cancers, including colorectal cancer, medullary thyroid carcinoma, and pheochromocytoma. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
