Specificity
Clone 3M19 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects RhoB. It targets an epitope within 17 amino acids from the C-terminal half.
Immunogen
KLH-conjugated linear peptide corresponding to 17 amino acids from the C-terminal half of human RhoB.
Application
Quality Control Testing
Evaluated by Western Blotting in MCF7 cell lysate.
Western Blotting Analysis: A 1:10,000 dilution of this antibody detected RhoB in MCF7 cell lysate.
Tested applications
Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected RhoB in HEK293 cells and 1:10,000 dilution detected RhoB in Rat Brain tissue lysates.
Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected RhoB in human cerebral cortex tissue sections.
Affinity Binding Assay: A representative lot of this antibody bound RhoB with a KD of 4.6 x 10-7 in an affinity binding assay.

Target description
Rho-related GTP-binding protein RhoB (UniProt: P62745; also known as Rho cDNA clone 6, h6) is encoded by the RHOB (also known as ARH6, ARHB) gene (Gene ID: 388) in human. RhoB is a member of the Ras super-family of small GTP-binding proteins that is synthesized with a short propeptide (aa 194-196), which is subsequently cleaved off to produce the mature active form. It has three nucleotide-binding regions (aa 12-19; 59-63; and 117-120). It is involved in hydrolysis of GTP and is active in GTP-bound form. RhoB is a short-lived Rho GTPase whose expression is inducible by a variety of stimuli including growth factors and stress stimuli. Its levels are up-regulated by DNA damaging agents, such as hydrogen peroxide or ionizing radiation. It plays a negative role in tumorigenesis as its deletion is known to cause tumor formation. It is reported to mediate apoptosis in neoplastically transformed cells following DNA damage. It is not considered to be essential for development but affects cell adhesion and growth factor signaling in transformed cells. RhoB participates in sorting and degradation of growth factors and cytokine receptors. It is shown to be critical for the inflammatory response of endothelial cells to TNFa, possibly through MAP kinase activation downstream of the TNFR. RhoB displays 83% homology with RhoA. However, unlike RhoA, which is cytosolic and translocates to plasma membrane upon activation, RhoB localizes to endosomes/multivesicular bodies. Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.
Reconstitution
300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Storage and Stability
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
