Specificity
Clone 1C18 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects RUNX1/AML1. It targets an epitope within 22 amino acids from the internal region.
Immunogen
KLH-conjugated linear peptide corresponding to 22 amino acids from the internal region of human Runt-related transcription factor 1 (RUNX1).
Application
Quality Control Testing
Evaluated by Western Blotting in RAW264.7 cell lysate.
Western Blotting Analysis: A 1:1,000 dilution of this antibody detected RUNX1 in RAW264.7 cell lysate.
Tested Applications
Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected RUNX1 in Rat Thymus lysates.
Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected RUNX1 in Jurkat cells.
Affinity Binding Assay: A representative lot of this antibody bound RUNX1 peptide with a KD of 2.5 x 10-7 in an affinity binding assay.

Target description
Runt-related transcription factor 1 (UniProt: Q01196; also known as Acute myeloid leukemia 1 protein, Core-binding factor subunit alpha-2, CBF-alpha-2, Oncogene AML-1, Polyomavirus enhancer-binding protein 2 alpha B subunit, PEA2-alpha B, PEBP2-alpha B, SL3-3 enhancer factor 1 alpha B subunit, SL3/AKV core-binding factor alpha B subunit) is encoded by the RUNX1 (also known as AML1, CBFA2) gene (Gene ID: 861) in human. RUNX members are shown to modulate the transcription of their target genes through recognizing the core consensus binding sequence 5′-TGTGGT-3′. RUNX1 is a core-binding transcription factor that is reported to be essential for the establishment of definitive hematopoiesis in vertebrates. It is expressed in all tissues except brain and heart. Its highest levels are observed in thymus, bone marrow, and peripheral blood. RUNX1 forms the heterodimeric complex core-binding factor (CBF) with CBFB and controls the expression of its target genes involved in hematopoietic differentiation, ribosome biogenesis, cell cycle regulation, and p53 and transforming growth factor β signaling pathways. RUNX1 binds DNA as a monomer through its Runt domain (aa 50-178) and its DNA-binding is increased by heterodimerization. RUNX1 is phosphorylated on serine 249 and 276 and threonine 273 by homeodomain interacting protein kinase 2 (HIPK2) when associated with CBFB and DNA. This phosphorylation promotes subsequent EP300 phosphorylation. RUNX1 gene is one of the most frequently mutated genes in a variety of hematological malignancies. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.
Reconstitution
300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Storage and Stability
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
