Specificity
Clone 3M19 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects Synaptonemal complex protein 3 (SCP-3). It targets an epitope within 25 amino acids from the N-terminal region.
Immunogen
KLH-conjugated linear peptide corresponding to 25 amino acids from the N-terminal region of human Synaptonemal complex protein 3 (SCP-3).
Application
Quality Control Testing
Evaluated by Western Blotting in Human testis tissue lysate.
Western Blotting Analysis: A 1:1,000 dilution of this antibody detected SCP-3 in Human testis tissue lysate.
Tested Applications
Immunofluorescence Analysis: A 1:1,000 dilution from a representative lot detected SCP-3 in Human testis tissue sections.
Affinity Binding Assay: A representative lot of this antibody bound SCP-3 peptide with a KD of 1.6 x 10-8 in an affinity binding assay.
Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected SCP-3 in Human testis tissue sections.
Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected SCP-3 in NTERA-2 cells.

Target description
Synaptonemal complex protein 3 (UniProt: Q8IZU3; also known as SCP-3) is encoded by the SCP3 (also known as SYCP3) gene (Gene ID: 50511) in human. SCP-3 is a homotetrameric protein that is a component of the lateral elements of synaptonemal complex, a meiosis-specific protein structure essential for synapsis of homologous chromosomes. The tetrameric helix bundles assemble end to end into long homopolimeric fibers that exhibit a transversal striation with a periodicity of about 20 nm. It contains a central coiled-coil domain (aa 66-223) and a sequence in the C-terminal region (aa 231-236) that is important for its oligomerization and fiber formation. It is required for centromere pairing during meiosis in male germ cells and is essential for normal meiosis during spermatogenesis and male fertility. Male primordial germ cells are reported to express SCP-3 for a short time before undergoing G1 arrest. Overexpression of SCP-3 has been reported in various cancer cells and is associated with tumor stage and grade. Its overexpression promotes Akt-mediated tumorigenesis both in vivo and in vitro. Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
Reconstitution
300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Storage and Stability
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
