Specificity

Clone 1M8 is a ZooMAb^® Rabbit recombinant monoclonal antibody that specifically detects TAF6L. It targets an epitope within 17 amino acids from the N-terminal region.

Immunogen

KLH-conjugated linear peptide corresponding to 17 amino acids from the N-terminal region of human TAF6L.

Application

Quality Control Testing

Evaluated by Western Blotting in RAW264.7 cell lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected TAF6L in RAW264.7 cell lysate.

Tested applications

Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected TAF6L in Jurkat and NIH3T3 cell lysates.

Affinity Binding Assay: A representative lot of this antibody bound TAF6L with a KD of 4.4 x 10-8 in an affinity binding assay.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected TAF6L in human testis and human vaginal tissue sections.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected TAF6L in MCF7 cells.

Target description

TAF6-like RNA polymerase II p300/CBP-associated factor-associated factor 65 kDa subunit 6L (UniProt: Q9Y6J9; also known as TATA-box binding protein associated factor 6 like, PCAF-associated factor 65-alpha, PAF65-alpha, TAF6L) is encoded by the TAF6L (also known as PAF65A) gene (Gene ID: 10629) in human. TAF6L functions as a component of the PCAF complex that is composed of a number of TBP-associated factors, such as TAF5, TAF5L, TAF6, TAF6L, TAF9, TAF10 and TAF12. This complex is capable of efficiently acetylating histones in a nucleosomal context. TAF6L along with TAF5L, acts as an epigenetic regulator essential for somatic cell reprogramming. It regulates target genes through H3K9ac deposition and Myc recruitment which trigger Myc regulatory network to orchestrate gene expression programs to control embryonic stem cell state. It functions with Myc to activate target gene expression through RNA polymerase II pause release. This ZooMAb recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
