General description
Tubulin polymerization promoting protein (TPPP), also known as p25, is expressed in myelinating oligodendrocytes. TPPP is mapped to human chromosome 5p15.33. It has a p25 domain and is a disordered protein.

Biochem/physiol Actions
Tubulin polymerization promoting protein (TPPP) regulates oligodendrocyte differentiation. It interacts with tubulinand regulates microtubule bundle formation. Phosphorylation of TPPP at threonine or serine residue by mitogen-activated protein kinase 2 or by LIM kinase (LIMK) impactstubulin polymerization functionality. High expression of TPPP favors acetylation of microtubules. Deletion in the gene locus of TPPP may be implicated in the pathogenesis of cerebellar hypoplasia and facial dysmorphism in fetus. Mutations in the TPPP gene contributes to tumor progression in small cell lung cancer. TPPP localization with α-synuclein may help in the understanding neurodegeneration in Parkinson′s and Alzheimer′s diseases. TPPP promotes abnormal microtubule assemblies in alpha-synucleinopathies.
