General description
Wybutosine (yW) is a hypermodified guanosine at the 3-prime position adjacent to the anticodon of phenylalanine tRNA that stabilizes codon-anticodon interactions during decoding on the ribosome. TRMT12 is the human homolog of a yeast gene essential for yW synthesis (Noma and Suzuki, 2006).[supplied by OMIM

Biochem/physiol Actions
TRMT12 (tRNA methyltransferase 12 homolog) gene is amplified and over-expressed in breast cancer. This enzyme is responsible for catalyzing the third step in wybutosine biosynthetic pathway. In cancer, up-regulation of this gene leads to disruption in SMG1 protein function, which is a part of nonsense mediated decay pathway (NMD). Thus, it might contribute to the escape of damaged DNA from checkpoints.
