General description
Tryptophan hydroxylase (TPH) is an enzyme involved in the synthesis of the neurotransmitter serotonin. It is responsible for addition of the -OH group (hydroxylation) to the 5 position to form the amino acid 5-hydroxytryptophan (5-HTP), which is the initial and rate-limiting step in the synthesis of the hormone serotonin. It is also the first enzyme in the synthesis of melatonin.
Specificity
Reacts with human and rat brain tissues by immunoblotting and immunohistochemistry. Based on the relatively high degree of homology of tryptophan hydroxylase, the antibody should cross-react with other mammalian species.

Immunogen
Recombinant rabbit tryptophan hydroxylase, isolated as inclusion bodies from E. coli and purified by preparative SDS-PAGE.

Detect Tryptophan Hydroxylase using this Anti-Tryptophan Hydroxylase Antibody validated for use in IH & WB.
Immunohistochemistry(peroxidase):
Performed on formaldehyde fixed tissue of a previous lot. Useful for cell body but not fiber staining.
Optimal working dilutions must be determined by the end user.
Quality
Routinely evaluated by Western Blot on PC12 lysates.
Western Blot Analysis:
1:1000 dilution of this lot detected Tryptophan Hydroxylase on 10 μg of PC12 lysates.

Storage and Stability
Stable for 6 months at -20ºC to -80ºC in undiluted aliquots from date of receipt.
Handling Recommendations: Upon receipt, and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance. Note: Variability in freezer temperatures below -20°C may cause glycerol-containing solutions to become frozen during storage.
