Specificity
Clone 1B7 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects Virilizer. It targets an epitope within 19 amino acids from the C-terminal region.
Immunogen
KLH-conjugated linear peptide corresponding to 19 amino acids from the C-terminal region of human Virilizer/VIRMA.
Application
Quality Control Testing
Evaluated by Western Blotting in Raji cell nuclear extract.
Western Blotting Analysis: A 1:1,000 dilution of this antibody detected Virilizer/VIRMA in Raji cell nuclear extract.
Tested Applications
Affinity Binding Assay: A representative lot of this antibody bound Virilizer/VIRMA peptide with a KD of 6.5 x 10-7 in an affinity binding assay.
Immunocytochemistry Analysis: A 1:1,000 dilution from a representative lot detected Virilizer/VIRMA in MCF-7 cells.
Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected Virilizer/VIRMA in human testis tissue sections.
Western Blotting Analysis: A 1:5000 dilution of a representative lot of this antibody detected Virilizer/VIRMA in Hek293T and mouse embryonic stem cells.
Immunohistochemistry Analysis: A 1:250 dilution of a representative lot of this antibody detected Virilizer/VIRMA in Formalin Fixed Paraffin Embedded (FFPE) human testis and human renal cell carcinoma tissue sections.

Target description
Protein virilizer homolog (UniProt: Q69YN4; also known as VIRMA) is encoded by the VIRMA (also known as KIAA1429, MSTP054) gene (Gene ID: 25962) in human. Virilizer is a member of the vir family of proteins that is an associated component of the WMM complex. This complex mediates N6-methyladenosine (m6A) methylation of RNAs, a modification that plays a role in the efficiency of mRNA splicing and RNA processing. It acts as a key regulator of m6A methylation by promoting m6A methylation of mRNAs in the 3′-UTR near the stop codon. It is involved in the recruitment of the catalytic core components METTL3 and METTL14, thereby guiding m6A methylation at specific sites. It is mainly localized in the nucleus with some fraction also observed in the cytoplasm. In Drosophila, virilizer is shown to be essential for male and female viability and is required for the production of eggs capable of embryonic development. Four isoforms of virilizer/VIRMA have been described that are produced by alternative splicing. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.
Reconstitution
300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Storage and Stability
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
