Chondroitin Sulfate (CS) is a type of sulfated GAGs composed of disaccharide units (GlcUA β 1-3GalNAc) composed of glucuronic acid (GlcUA) and N-acetylgalactosamine (GalNAc), which are repeatedly linked by β -1,4-glycosidic bonds. In the process of biosynthesis, the chondroitin basic skeleton of CS (-4GlcA β 1-3GalNAc β 1-) is first synthesized on specific serine residues of the core protein. During this process, the C6 and C-4 of GalNAc and the C2 of GlcA undergo sulfation modifications in the sugar chain by various sulfate transferases, forming disaccharide units with multiple different and sulfation modes. CS from different animal tissue sources are usually named based on the main disaccharide units they contain or the rare disaccharide units they are rich in, such as CS-A from mammalian cartilage, which mainly contains the 4-O-sulfated disaccharide GlcA β 1-3GalNAc (4S) (A unit); The CS-C derived from shark cartilage mainly contains the 6-O-sulfated disaccharide GlcA β 1-3GalNAc (6S) (C unit); CS-D derived from shark wings is rich in rare 2,6-O-sulfated disaccharide GlcA (2S) β 1-3GalNAc (6S) (D unit); CS-E derived from squid cartilage is rich in rare 4,6-O-sulfated disaccharide GlcA β 1-3GalNAc (4S, 6S) (E unit).
