General description
This control is intended for use as a positive control for knockout of human gene targets when used with clones of the Sigma lenti CRISPRs. The DNA format of this control construct permits direct transfection in knockout experiments or use in the viral packaging process (for lentiviral-based knockdown experiments).
Validated CRISPR site, which serves as an experimental control for the Sigma lenti CRISPRs. A one-component positive control system consisting of a EF1-alpha-driven Cas9, a U6-driven guide RNA plasmid targeting the human EMX1 gene, with both Puromycin and GFP co-expressed with Cas9. The Sigma lenti CRISPR Human EMX1 Positive Control Vector is a lentiviral-based plasmid vector that contains a guide RNA sequence targeting the human EMX1 (empty spiracles homeobox 1) gene. EMX1 belongs to the EMX homeobox family. The vector backbone is the same as the Sigma lenti CRISPR products.

Features and Benefits
Serves as an experimental control for the CRISPR editing workflow using lenti CRISPRs. Allows for validation of your system with the lenti CRISPR/Cas9 system. A positive result in a mis-match detection assay will indicate validation of your system.
Physical form
Ampicillin and puromycin antibiotic resistance genes provide selection in bacterial or mammalian cells, respectively. Also, self-inactivating replication incompetent viral particles can be produced in packaging cells (HEK293T) by co-transfection with compatible packaging plasmids (Product No. SHP001). The human EMX1 positive Control Vector is provided as 1 ug of plasmid DNA in Tris-EDTA (TE).
