ddNTP Mix (10mM each, Suitable for MALDI-MS) is a mixture of ddATP, ddCTP, ddGTP and ddTTP at 10mM each, specifically designed for MALDI-TOF MS analysis. ddNTPs are Mass-Modified Terminators and can be used for mass spectrometry analysis after single base primer extension. The ddTTP is specially modified to have a larger molecular weight than regular ddTTP. ddATP, ddCTP, ddGTP and ddTTP after incorporation into DNA have distinguishable molecular weights of approximately 271, 247, 287, and 327 Da, respectively, facilitating high-throughput single nucleotide polymorphism (SNP) analysis by MassArray mass spectrometry.With the discovery of several matrices capable of ionizing DNA and RNA, MALDI-TOF MS has become a powerful toll for the study of nucleic acids. It can be used to detect sequence variations such as mutations, single nucleotide polymorphisms (SNPs), insertions/deletions, and alternative splicing, as well as quantitative changes of gene copy numbers, gene expression, and allelic expression. Additionally, it can also be used to detect the methylation of genomic DNA and post-transcriptional modifications of tRNA and rRNA [1].ddNTP (Dideoxynucleoside Triphosphate) contains a dideoxy ribose group and lacks a 3' OH group. Therefore, unlike dNTP, it cannot form a phosphodiester bond during DNA synthesis or amplification, leading to chain termination.The molecular weight difference between the products generated by ordinary ddNTP after single base primer extension ranges from 9-40 Da, making it difficult to perform high-throughput genotyping by mass spectrometry. The ddTTP in this product has been specially modified to have a larger molecular weight than regular ddTTP, resulting in a molecular weight difference of 15 Da or more in the products, which allows genotyping based on the mass difference of the terminal target base [2-3].
