General description
Fibronectin is composed of two cell adhesion domains namely, the central domain for cell-binding made of adhesion-recognizing tetrapeptide, and the alternatively spliced type III connecting segment (IIICS). It contains a tetrapeptide region, from amino acid 90-109, called CS5, which is essential for adhesion mediated by IIICS. Both CS5 and CS1 mediate melanoma cell spreading, when stimulated by immunoglobulin (Ig)G, with CS1 exhibiting higher activity.
Biochem/physiol Actions
Fibronectin Type III Connecting Segment (IIICS) is responsible for melanoma cell adhesion. It is the predominant binding site of integrin α4β1, which is mediated through the aspartic acid residue present at position 21. Studies in chicken show that alternative splicing in this region during development of the peripheral nervous system, is essential for the control cell-cell adhesion.
