These media are prepared by using HiVeg infusion and HiVeg hydrolysate No.1 which are free of BSE/TSE risks. Heart Infusion Agar is used for mass cultivation of organisms for preparation of vaccines. It can be used as a base for preparation of Blood Agar to determine haemolytic reactions. This medium was used for detection and enumeration of haemolytic Streptococci in milk (1). A liquid medium containing an infusion of meat was one of the first media used for the cultivation of bacteria. Many modifications have been done since then. The growth factors were described by Lloyd (2) and Cole and Lloyd (3). Huntoon showed that highly pathogenic microorganisms as Meningococci and Pneumococci grow in infusion medium without enrichment (4). The Heart Infusion Agar / Broth, HiVeg are equivalent to these media by replacing animal based peptone with vegetable peptone. HiVeg hydrolysate No.1 and HiVeg infusion provides nutritional requirements to pathogenic bacteria. Heart Infusion Broth, HiVeg can be modified by adding dextrose, blood or other ingredients for use in variety of purposes. The medium can also be used for the cultivation of some anaerobes by addition of 0.1 - 0.2% agar. The addition of 5% sheep blood provides additional growth factors and is used to determine haemolytic reactions. Addition of 65 g/l of sodium chloride will aid in differentiating Enterococci from other Streptococci, as the Enterococci grow in presence of high salt while other Streptococci as pyogenic, virindans and lactic are unable to grow. Directions: Suspend 40.0 grams of MV169 or 25.0 grams of MV170 in 1000 ml of distilled water. Heat to boiling to dissolve the medium completely. Sterilize by autoclaving at 15 lbs pressure (121°C) for 15 minutes. If desired 5% sterile defibrinated blood may be added. Mix well and pour into sterile petri plates or tubes.
