Cell Line Description
The HtTA-1 cell line was derived from HeLa cells being stably transfected with a tetracycline-controlled transactivator (tTA) consisting of the tet repressor fused with the activating domain of virion protein 16 of herpes simplex virus. This transactivator stimulates transcription from a minimal promoter sequence derived from the human cytomegalovirus promoter IE combined with tet operator sequences. DMEM + 2mM Glutamine + 10% Foetal Bovine Serum (FBS).
Subculture Routine
Split sub-confluent cultures (70-80%) 1:5 to 1:20 i.e. seeding at 2x1,000 to 2x10,000 cells/cm2 using 0.25% trypsin or trypsin/EDTA; 5% CO2; 37°C.
