IgM antibodies are present as pentamers in the serum and are produced in response to antigens
IgM from bovine serum is purified from normal bovine serum by precipitation and gel filtration techniques.
IgM is a highly conserved antibody in vertebrates and is expressed early during immune response. It exists as a pentamer and is secreted by peritoneal B cells.
Application
IgM from bovine serum has been used for screening the lipopolysaccharide levels in Limulus amoebocyte lysate (LAL) assay. IgM has been used in flow cytometry assay for detecting FcγRIV expression and immunoglobulin binding.
Purified bovine IgM may be used as a reference antigen, standard, blocking agent, or coating protein in a variety of immunoassays including ELISA, dot immunobinding, Western immunoblotting, immunodiffusion, and immunoelectrophoresis. Other applications include starting materials for the preparation of immunogens and solid phase immunoadsorbents.
Biochem/physiol Actions
IgM plays a key role in engulfing of apoptotic cells. A reduction in serum IgM levels leads to increased autoimmune response and higher risk for infections. IgM displays polyreactive and autoreactive functionality. It plays a key role in tissue homeostasis by mediating clearance of tissue based molecules.

Solution in 0.05 M Tris-HCl, 0.15 M sodium chloride and 0.01 M glycine, pH 7.4, containing 15 mM sodium azide
