Cell Line Origin
Human Caucasian colon adenocarcinoma
Cell Line Description
A trypsinised variant of LS180 (Sigma Catalogue number. 87021202), both lines being derived from a 58 year old female with Dukes type B adenocarcinoma of the colon. The cells have microvilli and intracytoplasmic vacuoles. The cells are tumorigenic in nude mice.
Application
LS174T is part of the three-dimensional (3D) heterotypic spheroid model developed to assess cancer immunotherapy agents in vitro. It has been used to study the interaction of enterohaemorrhagic Escherichia coli (EHEC) with the intestinal mucus layer produced by LS174T cells.
Culture Medium
EMEM (EBSS) + 2mM Glutamine + 1% Non Essential Amino Acids (NEAA) + 10% Foetal Bovine Serum (FBS).
Subculture Routine
Split sub-confluent cultures (70-80%) 1:2 to 1:5 i.e. seeding at 2-5x10,000 cells/cm2 using 0.25% trypsin or trypsin/EDTA; 5% CO2; 37°C May take up to 7 days for cells to attach. Cells grow in islands and tend to pile on top of each other. Some cells ma
