Cell Line Description
The WT1-expressing mesonephric cell line M15 (alias Meso15) was established from mouse mesonephros transgenically expressing the large T protein of polyoma virus under the control of the early viral enhancer. As a tumour suppresser gene with a key role in urogenital development, WT1 is implicated as predisposition gene in the pathogenesis of Wilms′ tumour (WT).
Application
Gene expression and protein studies connected to kidney development and Wilms′ tumourigenesis.
Culture Medium
DMEM + 2mM Glutamine + 10% Foetal Bovine Serum (FBS).
Subculture Routine
Split sub-confluent cultures (70-80%) 1:5 to 1:10 i.e. seeding at 5x1,000 to 1x10,000 cells/cm2 using 0.25% trypsin or trypsin/EDTA; 5% CO2; 37°C. NB: Cell growth is impaired at lower densities.
