Cell Line Description
The cell line PEA-10 is heterozygous for a disruption of the gene for low density lipoprotein receptor-related protein (LRP). PEA-10 was derived from fibroblasts from day 15 C57BL/6x129 F1 hybrid mouse embryos. Together with the cell lines PEA-13 (Sigma Catalogue number. 97040311) and MEF-1 (Sigma Catalogue number. 98061101) an experimental system is available for the analysis of cellular uptake of functionally diverse ligands and the effect of LRP deficiency. This is of importance since LRP deficient mouse embryos die early during gestation.
Application
Study of LRP and LRP deficiency.
Culture Medium
DMEM + 2mM Glutamine + 10% Foetal Bovine Serum (FBS).
Subculture Routine
Split confleunt cultures 1:6 to 1:10 i.e. seeding 2-4 x 10,000 cells/cm2 using 0.25% trypsin or trypsin/EDTA; 5% CO2; 37°C
