Recombinant ubiquitin fused at the N-terminus to GST and expressed in E. coli. Useful for glutathione affinity purification of ubiquitinated molecules. Can also be used for immunodetection of conjugates using GST antibodies. Facilitates the visualization of polyUb chains due to larger ladder intervals.
Application
Ubiquitin, GST-Fusion, Recombinant, E. coli has been used for in vivo and in vitro ubiquitination assays, to study the role of FBXL2-Grp94-EGFR axis in non-small cell lung cancer (NSCLC) development and the role of F-box protein 3 (FBXO3) in promotion of breast cancer metastasis.
Biochem/physiol Actions
The ubiquitin (Ub) system plays a crucial role in protein homeostasis by maintaining protein balance in pathways related to DNA repair, cell cycle progression, apoptosis (programmed cell death), receptor-mediated endocytosis, and signal transduction. Imbalances or dysfunctions within this system can lead to pathological conditions. Apart from this, disruptions in ubiquitin pathways can also contribute to the development of human diseases, including neurodegenerative conditions and tumors.
