YO-PRO-1 (YP1), also known as oxazole yellow, is a non-permeable green fluorescent DNA dye which cannot pass through the biologically active cell plasma membrane of healthy cells, but can pass through the disordered cell membrane of apoptotic and necrotic cells to reach the nucleus. In nucleus, YP1 binds with DNA and emits bright green fluorescence while YP1 in free state has no fluorescence. YP1 can stain nuclei of both apoptic and necrotic cells in green. However PI only stains nuclei of necrotic cells and emits bright red fluorescence when binding to DNA. Therefore, YP1 is often used in combination with PI to differentiate apoptosis from necrosis.YP1 has the molecular formula of C24H29I2N3O with a molecular weight of 629.32. Its CAS number is 152068-09-2. YP1 binding with DNA has an emission maximum at 509nm, when excited at 491nm. Please see Figure 1 for the excitation and emission spectra of YP1 when binding to DNA. It provides comparable accuracy and sensitivity in apoptosis detection to the Annexin V-FITC probe that is conventionally used for the detection of phosphatidylserine exposure in apoptotic cells.This product is sterile and provided in 1mM stock solution. Dilute this product at a ratio of 1Precautions：Fluorescence intensity decays. Fluorescence observation should be completed within 1 hour after staining. Stained samples to be observed should be kept on ice in the dark.When using the YP1 at the first time, make aliquots and store at -20ºC in the dark to avoid repeated freeze-thaws.YP1 is irritating.
