Acrylamide and bis-acrylamide are major ingredients in polyacrylamide gels, a commonly used matrix for electrophoretic separation of nucleic acids and proteins. Fragments of DNA produced by various molecular biology techniques (eg. restriction endonuclease digestion, dideoxy sequencing, or the Maxam and Gilbert sequencing methods) can be separated by polyacrylamide gel electrophoresis (PAGE). Proteins may also be separated with polyacrylamide gels (eg. denaturing gels using SDS or urea, isoelectric focusing gels, native gels etc). Ready-to-use acrylamide:bis-acrylamide solutions are convenient in avoiding the risks associated with handling the powder. The reagents are accurately pre-blended to produce a 40% (w/v) stock solution. The concentration is based on the total weight of both the acrylamide and bis-acrylamide (29:1).
