General description

Specificity

Clone 1E4 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects DICER1. It targets an epitope within 20 amino acids from the internal region.

Immunogen

KLH-conjugated linear peptide corresponding to 20 amino acids from the internal region of human endoribonuclease Dicer (DICER1).

Application

Quality Control Testing

Evaluated by Western Blotting in HeLa cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected DICER1 in HeLa cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected DICER1 in A431 cell lysate.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected DICER1 in Human small intestine tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound DICER1 peptide with a KD of 5.0 x 10-7 in an affinity binding assay.

Target description

Endoribonuclease Dicer (UniProt: Q9UPY3; also known as EC:3.1.26.3, Helicase with RNase motif, Helicase MOI) is encoded by the DICER1 (also known as DICER, HERNA, KIAA0928) gene (Gene ID: 23405) in human. DICER is a multidomain endonuclease that belongs to the fourth class of type III ribonuclease (RNase III) enzymes. It is a component of the RNA-induced silencing complex (RISC) that is composed of DICER1, AGO2 and TARBP2. Here DICER1 and TARBP2 are shown to be essential in processing precursor microRNAs (miRNA) into mature miRNAs and then load them onto AGO2. The processed mature RNAs are subsequently incorporated into RISC. DICER cleaves naturally occurring LONG® dsRNAs and short hairpin pre-miRNA into fragments of twenty-one to twenty-three nucleotides with 3′ overhang of two nucleotides, producing respectively short interfering RNAs (siRNA) and mature microRNAs that serve as a guide to direct RISC to complementary RNAs to degrade them or prevent their translation. DICER expression is tightly regulated and its inactivation results in loss of mature miRNAs and accumulation of miRNA precursors. Its dysfunctions and abnormal miRNA processing have been linked to aging and various neurodegenerative diseases. Its levels are downregulated in patients suffering from age-related diseases, such as Parkinson€²s disease and amyotrophic lateral sclerosis (ALS). Mutations in DICER1 gene have been linked to Pleuropulmonary blastoma, a rare pediatric intrathoracic neoplasm that arises during fetal lung development. DICER1 consists of an N-terminal helicase, a platform-PAZ-connector Helix domain (aa 891-1042), two RNASE III domains (aa 1276-1403 and 1666-1824) and a C-terminal double-stranded RNA-binding domain (aa 1849-1914). Three isoforms of DICER1 have been described that are produced by alternative splicing. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, Affinity, reproducibility, and stability over conventional monoclonals. Reconstitution

300 g/mL after reconstitution at 25 L per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 L of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
