General description

Tumor necrosis factor receptor superfamily member 16 (UniProt P07174; also known as CD271, Gp80-LNGFR, Low-affinity nerve growth factor receptor, Low affinity neurotrophin receptor p75NTR, NGF receptor, p75 ICD) is encoded by the Ngfr (also known as Tnfrsf16) gene (Gene ID 24596) in rat species. p75NTR is a single pass type I membrane protein that can bind to NGF, BDNF, NT-3 and NT-4 and mediate neuronal cell survival and death. NGF and p75NTR are considered as important mediators of pain. Levels of p75NTR are shown to increase following nerve injury and its functional inhibition suppresses injury-induced neuropathic pain in rats. In non-neural cells, p75NTR expression correlates with differentiation and dedifferentiation processes and it is transient expression is observed during development of kidney, lungs, ovary, and muscle. Down-regulation of p75NTR is considered to be important during myogenic differentiation and muscle repair. Ref.: Fukui, Y. et al. (2010). J. Orthoped. Res. 28, 279-283. Deponti, D. et al. (2009). Mol. Biol. Cell 20, 3620-3627.

Specificity

This antiserum detected the ~75 kDa full-length p75NTR (neurotrophin receptor) as well as the ~30 kDa CTF and the ~25 kDa ICD cleavage fragments by immunoprecipitation and Western blotting (Kenchappa, R.S., et al. (2006). Neuron. 50(2):219-232).

Immunogen

GST-tagged recombinant rat p75NTR ICD fragment.

Application

Anti-p75NTR, ICD, Cat. No. ABN1655, is a highly specific rabbit polyclonal antiserum that targets up75NTR (neurotrophin receptor) and has been tested in Immunocytochemistry, Immunofluorescence, Immunoprecipitation, and Western Blotting.

Immunocytochemistry Analysis: A representative lot immunostained 4% paraformaldehyde-fixed, 0.1% Triton X-100-permeabilized rat sympathetic neurons by fluorescent immunocytochemistry. Upregulated staining was observed in neurons infected with p75NTR ICD-expressing adenovirus (Kenchappa, R.S., et al. (2006). Neuron. 50(2):219-232).

Immunofluorescence Analysis: A representative lot localized p75ICD immunoreactivity in cryosections from various stages of chick embryos fixed with 4% paraformaldehyde and permeabilized by 0.1% Triton X-100 (López-Sánchez, N., et al. (2007). Physiol. Genomics. 30(2):156-171).

Immunoprecipitation Analysis: A representative lot co-immunoprecipitated NRIF with p75 from rat sympathetic neurons. Co-immunoprecipitation of exogenously expressed NRIF and p75 by this antiserum was also seen using 293 transfectant. Increased CTF and ICD fragments were immunoprecipitated from PMA-treated 293 transfectants and from BDNF-treated neurons (in the presence of proteasome inhibitor). Co-treatment with gamma-secretase inhibitor abolished ICD, but not CTF cleavage in PMA- and BDNF-treated cells (Kenchappa, R.S., et al. (2006). Neuron. 50(2):219-232).

Western Blotting Analysis: A representative lot detected p75 in rat sympathetic neurons. Increased ~30 kDa CTF and ~25 kDa ICD fragments were detected from BDNF- or PMA-, but not NGF-, treated neurons (in the presence of proteasome inhibitor). Co-treatment with gamma-secretase inhibitor abolished ICD, but not CTF cleavage in BDNF-treated cells (Kenchappa, R.S., et al. (2006). Neuron. 50(2):219-232).

Western Blotting Analysis: A representative lot detected p75 in superior cervical ganglia (SCG) whole tissue lysate from P4 and P24 postnatal rats. CTF and ICD fragments were detected only in P4, but not P24 SCG lysate (Kenchappa, R.S., et al. (2006). Neuron. 50(2):219-232).

Quality

Evaluated by Western Blotting in mouse retina tissue lysate.

Western Blotting Analysis: A 1:5,000 dilution of this antiserum detected the ~25 kDa p75NTR (neurotrophin receptor) ICD fragment in 10 µg of mouse retina tissue lysate.

Target description

~25 kDa observed (mouse p75 ICD). 44.65/42.71 kDa (chicken p75 with/without signal peptide; UniProt P18519), 44.69/42.42 kDa (mouse p75 with/without signal peptide; UniProt Q9Z0W1), 45.43/42.48 kDa (rat p75 with/without signal peptide; UniProt P07174). ~75/30/25 kDa (rat p75 full-length/CTF/ICD) reported (Kenchappa, R.S., et al. (2006). Neuron. 50(2):219-232). Uncharacterized bands may be observed in some lysate(s).

Physical form

Rabbit polyclonal antibody serum with 0.05% sodium azide.

Unpurified.

Storage and Stability

Stable for 1 year at -20°C from date of receipt.

Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
