Terrific Broth is a rich medium for cultivation of E. coli and is often used for growth to high density before plasmid purification. Tryptone and yeast extract provide nutrients, glycerol provides the carbon and energy source, and phosphate buffer helps stablize pH to prevent cell death. 1X Terrific Broth contains 1.2% (12 g/L) tryptone, 2.4% (24 g/L) yeast extract, 0.4% glycerol. Terrific Broth Complete also contains 17 mM monopotassium phosphate and 72 mM dipotassium phosphate. Some part numbers provide 10X phosphate buffer separately.
