The UltraBio¢"¢ Blocking Buffer has the following advantages. A. Fast and efficientPrecautions:Use flat-tip tweezers to gently handle PVDF and NC membranes on the corners or edges to avoid producing any scrapes, creases or indentations on the surface of membrane.PVDF membrane, once infiltrated and activated, should be kept wet by placing the membrane in Western transfer buffer or washing buffer according to the Western Blot procedures. Otherwise, an abnormal background that is hard to be blocked may be produced.To enable the diluted antibodies to be reused multiple times, the primary/secondary antibody dilutions should be stored at 4oC immediately after each use.Usually, it takes 5-15 minutes at room temperature to block PVDF or NC membranes. Use the buffers provided in this set at appropriate steps. 1. Usage of QuickBlock¢"¢ Blocking Buffer:a. After completing the protein transfer, wash the membrane with the 1X Washing Buffer for 1-2 minutes.b. Based on the size of the membrane, add a sufficient volume of QuickBlock¢"¢ Blocking Buffer in a petri dish or other appropriate container. For regular Western Blot, use 10ml of Blocking Buffer for a membrane of 6.6—8.5cm.c. Clamp one corner of the membrane with a flat-tip tweezer and place the membrane in QuickBlock¢"¢ Blocking Buffer for Western Blot. Make sure that the membrane is covered completely by the Blocking Buffer. Incubate for 10 minutes with agitation on a horizontal shaker. The incubation time can be from 5 to 15 minutes. Generally, 10 minutes of blocking can achieve better blocking results than 1 hour's blocking with BSA.2. Usage of QuickBlock¢"¢ Primary Antibody Dilution Buffer:a. Refer to the instructions for the primary antibody used, dilute the primary antibody with the QuickBlock¢"¢ Primary Antibody Dilution Buffer at an appropriate ratio (1:1000 or 1:500, etc), depending on the amount of target proteins in the sample. b. Add sufficient primary antibody dilutions to cover the membrane for incubation. c. After incubation, recover the primary antibody dilutions and store at 4oC immediately for later use.3. Usage of QuickBlock¢"¢ Secondary Antibody Dilution Buffer:a. Refer to the instructions for the secondary antibody used, dilute the secondary antibody with the QuickBlock¢"¢ Secondary Antibody Dilution Buffer at an appropriate ratio (1:1000 or 1:500, etc), depending on the quality of primary antibody used and the amount of target proteins in the sample. b. Add sufficient secondary antibody dilutions to cover the membrane for incubation. c. After incubation, recover the secondary antibody dilutions and store at 4oC immediately for later use.4. Usage of Washing Buffer (10X):a. Dilute the Washing Buffer (10X) with distilled H2O to 1X. The 1X washing buffer is stable at room temperature and should be used within 1-2 weeks. When turned turbid or precipitation occurs, the 1X washing buffer should be discarded.b. After incubation with primary and secondary antibodies, wash the membrane 3 times with sufficient 1X washing buffer for 5 minutes each. Wash more times or prolong the washing time if there is high background noise, which usually does not affect the Western results.
